A new method for the determination of aminopeptidase P activity using Gly-Pro-Hyp as the substrate was described. Enzymatically formed Pro-Hyp from the substrate in the assay mixture was chromatographically separated by high performance liquid chromatography (HPLC) and determined the absorbance of the peptide at 210 nm.
