内容記述 Bone morphogenetic protein (BMP) is an important bioactive protein, but its bioactivity depends upon the carrier. Therefore, many researchers have tested a considerable number of biomaterials as possible carriers. Squalane is used as an important base for various kind of cosmetics. Thus, it was considered that squalane might be a useful carrier of BMP, and consequently evaluated it histopathologically. Firstly to determine the safety of squalane fluid applied inside the body, the tissue reaction to subcutaneously embedded squalane fluid in adult SD rats was examined. The results are as follows: No macroscopic abnormalities were found in any part of the body of the experimental rats during the experimental period when compared with the control rats. Histopathologically, granulation tissue formation with slight inflammatory cell infiltration was elicited by the squalane fluid, although neither degeneration nor necrosis were recognized in the embedded area. Some of the squalane had been phagocytosed by macrophages in the granulation tissue, but most of it appeared to have been resorbed during the experimental period; no evidence of encapsulation was observed. Then, gelatin capsules containing squalane/BMP (partially purified) mixture were placed under the perimuscular membrane of ddY mice. Two kind of control, gelatin capsules containing only BMP (partially purified) and those bearing squalane only, were used. The embedded areas were histopathologically examined at 1 to 4 weeks after the operation. Varying amount of new bone were formed by endochondral ossification at 2 weeks. The bone of 3 and 4 week-specimens revealed bone marrow already formed. The observations revealed that the squalane/BMP (partially purified) mixture as well as BMP (partiallly purified) only control specimens elicited wide heterotopic bone formation with bone marrow tissue, although there was no bone formation in the squalane-only control group. Accordingly these data suggest that squalane fluid is safe for use inside the body, and that squalane is a useful fluid carrier of BMP: squalane/BMP mixture can be used as an osteoinductive biomaterial.
