内容記述 The tissue reactions to a root canal filling material and transplanted teeth through the experimental models were studied. Tooth columns were made from lower incisors which were removed from ten-week-old Wistar rats anesthetized. Canals of the columns were enlarged and filled with calcium-hydroxide paste mixed with silicone-oil. These tooth columns and removed teeth for control were autotransplanted into subcutaneous tissues. At 3 to 28 days after the experiment, these materials with surrounding tissues were removed and examined histopathologically. Results are as follows: Granulation tissue with macrophages and multinuclear giant cells (MGCs) proliferated around the paste after 7 days. MGCs had already appeared close to the cut surfaces of the dentin mimicking the root apecies at 7 days, while there were few MGCs in the controls at this stage. After 14 days, MGCs formed resorption lacunae on dentin surfaces both in the experiments and the controls. Furthermore, in the experimental specimens at 28 days, a MGC was observed lying on both the dentin and the paste. In the control specimens at the same days, tooth pieces with resorption lacunae were encapsulated by thin fibrous tissue. In conclusion, this experimental model is thought to be useful to investigate tissue reactions to filling paste and to clarify the behavior of MGCs, because our experiment seems to be able to bring the same histopathological changes as that of the periapical tissue after root canal filling.
